I have not actually tried this, but I expect it should "just work"(tm) as long as you install the optional server components to enable DirectX graphics and Store apps.
Have you tried this and encountered issues?

I have not tried this yet - but your statement is encouraging.
- Is there a command-line non-interactive command that can prepare a server for Win2D usage?
- Would these components likely be installed by default by any hosting provider (say Azure Websites)?
Potential usage context would be ASP.NET middleware for image processing.

I don't know a lot about Azure, but would be surprised if it had D3D graphics and Store app support / XAML UI stack enabled. You can install these components on a Windows Server SKU, but that is an OS upgrade operation that will require admin access.
