Familial Adenomatous Polyposis (FAP) is a rare inherited disorder carrying a 100% risk of colorectal cancer without proper intervention. Using publicly available data, we show that the APC deficiency causes global dysregulation in tissue patterning in addition to aberrant proliferation. Moreover, acquisition of genetic alterations during adenoma-carcinoma transition further exacerbates proliferative signaling and lineage diversity via cMYC-YAP driven transcriptional programming.
