The username should be simply username, then the domain argument should be used:
conn = SMBConnection(username='user_name',
password=password,
my_name='whatever',
remote_name='whatever',
domain='domain',
use_ntlm_v2=True, is_direct_tcp=True)
_conn = conn.connect('192.168.0.10', 445)
print(_conn)
