Make sure to deny the 'Muted' role the 'Send Messages' and 'Add Reactions' permission (this can be done easily with Beta's ))setup command). It should look something like this:

You also need to make sure that the 'roster' role does not have the 'Send Messages' and 'Add Reactions' permission enabled in that channel. Otherwise, these permissions will overlap and they will still be able to send messages and add reactions.

If this is a private channel only accessible by the role, you can fix this by giving the 'roster' role 'Read Messages' but leave 'Send Messages' and 'Add Reactions' to their default setting.
