I assume nvidia-smi works fine and lists the GPU? Try running https://gist.github.com/f0k/63a664160d016a491b2cbea15913d549, or compiling and running https://gist.github.com/f0k/0d6431e3faa60bffc788f8b4daa029b1. If they also fail, something is wrong with your CUDA installation. In the early days, I had to run the latter program as root once after booting to create a missing /dev entry, but I think this problem disappeared by now.

You were probably right because after un-installing and re-installing everything it's now working.
